Penn-Lewis was converted to Christianity at the age of sixteen. She became active in various ministries, including mission work, preaching, and teaching. In 1884, she married William Lewis and later worked alongside him in a number of evangelistic campaigns. Penn-Lewis was a prolific writer and speaker and was known for her insight into the deeper aspects of the Christian life.
 
Her writings focused on the spiritual warfare between the forces of good and evil and the importance of surrendering our lives to God. She wrote a number of books, including Soul and Spirit, War on the Saints, The Overcoming Life, and The Cross and the Coming of the Lord.
 
Penn-Lewis was an influential figure in the early Pentecostal movement and an advocate for the baptism of the Holy Spirit. She was also involved in various interdenominational conferences and was a key figure in the Keswick Convention movement. Penn-Lewis was a strong advocate for spiritual renewal and revival within the church. She believed that true revival could only come through prayer and a renewed commitment to following Jesus Christ.
